Ductile extension of 3-D external circumferential cracks in pipe structures

摘要: This study investigates the ductile fracture resistance of 3-D external circumferential cracks in the wall of a steel pipe under remote tension, using a damage-mechanism model originally proposed by Gurson and Tvergaard. The ductile crack extension utilizes an element extinction technique implemented in the computational cell framework. The key parameter for the computational cell method, i.e., the initial porosity ratio , is calibrated using both the fracture resistance and the load-deformation responses obtained from fracture tests of multiple single-edge bend [SE(B)] specimens made of high-strength steel, HY80, which has a yield strength of 630 MPa. The fracture resistance along the 3-D semi-elliptical crack front is computed from the calibrated cell model. Based on the similarity concept in the near-tip stress-strain fields, this study demonstrates that an equivalent 2-D axi-symmetric model provides conservative estimations of the fracture resistance for 3-D circumferential cracks in pipes.

Novel mutation c.1210-3C>G in with a poly-T tract of 5T affects mRNA splicing in a Chinese patient

摘要: Cystic fibrosis (CF) is a rare autosomal recessive disease with only one pathogenic gene cystic fibrosis transmembrane conductance regulator (CFTR). To identify the potential pathogenic mutations in a Chinese patient with CF, we conducted Sanger sequencing on the genomic DNA of the patient and his parents and detected all 27 coding exons of CFTR and their flanking intronic regions. The patient is a compound heterozygote of c.2909G>A, p.Gly970Asp in exon 18 and c.1210-3C>G in cis with a poly-T of 5T (T5) sequence, 3 bp upstream in intron 9. The splicing effect of c.1210-3C>G was verified via minigene assay in vitro, indicating that wild-type plasmid containing c.1210-3C together with T7 sequence produced a normal transcript and partial exon 10-skipping-transcript, whereas mutant plasmid containing c.1210-3G in cis with T5 sequence caused almost all mRNA to skip exon 10. Overall, c.1210-3C>G, the newly identified pathogenic mutation in our patient, in combination with T5 sequence in cis, affects the CFTR gene splicing and produces nearly no normal transcript in vitro. Moreover, this patient carries a p.Gly970Asp mutation, thus confirming the high-frequency of this mutation in Chinese patients with CF.

Ensemble unit and AI techniques for prediction of rock strain

摘要: The behavior of rock masses is influenced by a variety of forces, with measurement of stress and strain playing the most critical roles in assessing deformation. The laboratory test for determining strain at each location within rock samples is expensive and difficult but rock strain data are important for predicting failure of rock material. Many researchers employ AI technology in order to solve these difficulties. AI algorithms such as gradient boosting machine (GBM), support vector regression (SVR), random forest (RF), and group method of data handling (GMDH) are used to efficiently estimate the strain at every point within a rock sample. Additionally, the ensemble unit (EnU) may be utilized to evaluate rock strain. In this study, 3000 experimental data are used for the purpose of prediction. The obtained strain values are then evaluated using various statistical parameters and compared to each other using EnU. Ranking analysis, stress-strain curve, Young’s modulus, Poisson’s ratio, actual vs. predicted curve, error matrix and the Akaike’s information criterion (AIC) values are used for comparing models. The GBM model achieved 98.16% and 99.98% prediction accuracy (in terms of values of R2) in the longitudinal and lateral dimensions, respectively, during the testing phase. The GBM model, based on the experimental data, has the potential to be a new option for engineers to use when assessing rock strain.

Sustainable intensification of agriculture in Africa

摘要: Sustainable intensification is a key component of agricultural development in Africa, urgently needed to wean the continent off foreign food supply and to limit agricultural farmland expansion. It is expected that a relatively small fraction of farmers will adopt fertilizer technology, as profits in current economic settings are relatively small while risks are considerable with varying prices and uncertain yield responses. Many smallholders depend on off-farm income and local markets for food supply. Structural adjustments are therefore needed to allow management of larger units of land by trained farmers willing to take this opportunity, while recognizing land right sensitivities. There are large opportunities for African commodity crops to improve food security, including cassava and East African highland banana that strongly respond to fertilizer with limited environmental risks under good management. This requires investments in better functioning markets, local fertilizer production facilities that can produce regional crop blends and cost-efficient distribution networks, providing balanced fertilizers for African farmers.

摘要: Meta-analysis was used to determine whether maintenance intravesical bacillus Calmette-Guerin (BCG) could reduce recurrence after transurethral resection of tumor 1 grade 3 (T1G3) superficial bladder cancer. All available published data of randomized clinical trials comparing transurethral resection plus intravesical BCG to either resection alone or resection plus another treatment on the treatment results in patients with superficial bladder cancer of T1G3 were selected for analysis. Both the fixed effects model and random effects model were applied, and the odds ratio () with its 95% confidence interval (CI) was used as the effect size estimate. Sensitivity analysis and publication bias determination were performed by funnel plots and comparing s of different models. Within the follow-up period, 375 of 915 (41.0%) BCG-treated patients and 332 of 733 (45.3%) non-BCG-treated patients developed tumor recurrence. In the combined results, a statistically significant difference in the s for tumor recurrence between the two treatment groups was found (randomized model combined effect 0.58, 95% CI 0.41 to 0.83, = 0.003). The stratified meta-analysis did not show any statistically significant confounding effects on the results when stratified by BCG strains. The randomized model combined effect of Pasteur F and other strains were 0.50 (95% CI 0.26 to 0.95, = 0.04) and 0.63 (95% CI 0.40 to 0.99, = 0.04), respectively. Therefore, we came to the conclusion that adjuvant maintenance instillation BCG combined with transurethral resection of bladder tumor (TURBT) is an effective conservative treatment for preventing recurrence of T1G3 bladder cancer.

REINTEGRATION OF CROP-LIVESTOCK SYSTEMS IN EUROPE: AN OVERVIEW

摘要: Ongoing specialization of crop and livestock systems provides socioeconomic benefits to the farmer but has led to greater externalization of environmental costs when compared to mixed farming systems. Better integration of crop and livestock systems offers great potential to rebalance the economic and environmental trade-offs in both systems. The aims of this study were to analyze changes in farm structure and review and evaluate the potential for reintegrating specialized intensive crop and livestock systems, with specific emphasis on identifying the co-benefits and barriers to reintegration. Historically, animals were essential to recycle nutrients in the farming system but this became less important with the availability of synthetic fertilisers. Although mixed farm systems can be economically attractive, benefits of scale combined with socio-economic factors have resulted in on-farm and regional specialization with negative environmental impacts. Reintegration is therefore needed to reduce nutrient surpluses at farm, regional and national levels, and to improve soil quality in intensive cropping systems. Reintegration offers practical and cost-effective options to widen crop rotations and promotes the use of organic inputs and associated benefits, reducing dependency on synthetic fertilisers, biocides and manure processing costs. Circular agriculture goes beyond manure management and requires adaptation of both food production and consumption patterns, matching local capacity to produce with food demand. Consequently, feed transport, greenhouse gas emissions, nutrient surpluses and nutrient losses to the environment can be reduced. It is concluded that reintegration of specialized farms within a region can provide benefits to farmers but may also lead to further intensification of land use. New approaches within a food system context offer alternatives for reintegration, but require strong policy incentives which show clear, tangible and lasting benefits for farmers, the environment and the wider community.

A multi-scale model for CO

摘要: This paper presents a multi-scale model to simulate the multicomponent gas diffusion and flow in bulk coals for CO sequestration enhanced coalbed methane recovery. The model is developed based on a bi-dispersed structure model by assuming that coal consists of microporous micro-particles, meso/macro-pores and open microfractures. The bi-disperse diffusion theory and the Maxwell-Stefan approach were incorporated in the model, providing an improved simulation of the CH —CO /CH —N counter diffusion dynamics. In the model, the counter diffusion process is numerically coupled with the flow of the mixture gases occurring within macro-pores or fractures in coal so as to account for the interaction between diffusion and flow in gas transport through coals. The model was validated by both experimental data from literature and our CO flush tests, and shows an excellent agreement with the experiments. The results reveal that the gas diffusivities, in particular the micro-pore diffusivities are strongly concentration-dependent.

增强对流输注 (CED) 是一项颇具前景的技术,其借助压力驱动流来增强输注药物进入细胞间隙的穿透力。为进一步提升CED的药物分布容积,笔者发明了一种可产生局部亚致死热量的光纤微针装置。本文试图在琼脂糖组织模型中对该技术进行定量表征。在15 °C、20 °C、25 °C和30 °C的恒温条件下,染料的输注在质量分数为0.6%的琼脂糖组织模型中进行分析。输注指标通过自定义阴影成像技术和图像处理算法进行定量。利用所获数据构建一个分布容积的经验预测时序模型作为组织模型温度的函数。接下来通过一组概念验证实验来评估液体输注时新型光纤装置产生局部光加热的能力。恒温输注显示温度和分布容积呈正相关,在100 min时,在30 °C恒温条件下体积扩散是在15 °C恒温条件下的7倍。在光加热 (1064 nm,500 mW) 过程中,输注呈现相似的效果:与对照组 (0 mW) 相比,输注体积在4 h时增大了3.5倍。本文的分析和结果为体积扩散的热介导增强提供了特征描述及新思路。

State-of-the-art of intelligent minimally invasive surgical robots

摘要: A number of developed countries are rapidly turning into super-aged societies. Consequently, the demand for reduced surgical invasiveness and enhanced efficiency in the medical field has increased due to the need to reduce the physical burden on older patients and shorten their recovery period. Intelligent surgical robot systems offer high precision, high safety, and reduced invasiveness. This paper presents a review of current intelligent surgical robot systems. The history of robots and three types of intelligent surgical robots are discussed. The problems with current surgical robot systems are then analyzed. Several aspects that should be considered in designing new surgical systems are discussed in detail. The paper ends with a summary of the work and a discussion of future prospects for surgical robot development.

HVAC系统的模糊预测函数控制器设计

摘要:

针对暖通空调HVAC系统中由于存在高度非线性、时变特征以及扰动和不确定性等因素而难以控制的特点,提出基于Takagi-Sugeno(T-S)模糊模型的预测函数控制器设计方法。该方法通过最小二乘辨识算法建立系统的模糊T-S模型,然后基于模糊全局线性化预测模型,采用预测函数控制算法设计系统控制律。仿真实验结果表明该算法是一种跟踪性能好、鲁棒性强的有效控制方法。

R158Q and G212S, novel pathogenic compound heterozygous variants in of Gitelman syndrome

摘要: The dysfunction of Na+-Cl cotransporter (NCC) caused by mutations in solute carrier family12, member 3 gene (SLC12A3) primarily causes Gitelman syndrome (GS). In identifying the pathogenicity of R158Q and G212S variants of SLC12A3, we evaluated the pathogenicity by bioinformatic, expression, and localization analysis of two variants from a patient in our cohort. The prediction of mutant protein showed that p.R158Q and p.G212S could alter protein’s three-dimensional structure. Western blot showed a decrease of mutant Ncc. Immunofluorescence of the two mutations revealed a diffuse positive staining below the plasma membrane. Meanwhile, we conducted a compound heterozygous model—Ncc R156Q/G210S mice corresponding to human NCC R158Q/G212S. NccR156Q/G210S mice clearly exhibited typical GS features, including hypokalemia, hypomagnesemia, and increased fractional excretion of K+ and Mg2+ with a normal blood pressure level, which made NccR156Q/G210S mice an optimal mouse model for further study of GS. A dramatic decrease and abnormal localization of the mutant Ncc in distal convoluted tubules contributed to the phenotype. The hydrochlorothiazide test showed a loss of function of mutant Ncc in NccR156Q/G210S mice. These findings indicated that R158Q and G212S variants of SLC12A3 were pathogenic variants of GS.

Simultaneous saccharification and fermentation of wheat bran flour into ethanol using coculture of amylotic

摘要: Studies on simultaneous saccharification and fermentation (SSF) of wheat bran flour, a grain milling residue as the substrate using coculture method were carried out with strains of starch digesting and nonstarch digesting and sugar fermenting in batch fermentation. Experiments based on central composite design (CCD) were conducted to maximize the glucose yield and to study the effects of substrate concentration, pH, temperature, and enzyme concentration on percentage conversion of wheat bran flour starch to glucose by treatment with fungal α-amylase and the above parameters were optimized using response surface methodology (RSM). The optimum values of substrate concentration, pH, temperature, and enzyme concentration were found to be 200 g/L, 5.5, 65°C and 7.5 IU, respectively, in the starch saccharification step. The effects of pH, temperature and substrate concentration on ethanol concentration, biomass and reducing sugar concentration were also investigated. The optimum temperature and pH were found to be 30°C and 5.5, respectively. The wheat bran flour solution equivalent to 6% ( / ) initial starch concentration gave the highest ethanol concentration of 23.1 g/L after 48 h of fermentation at optimum conditions of pH and temperature. The growth kinetics was modeled using Monod model and Logistic model and product formation kinetics using Leudeking-Piret model. Simultaneous saccharificiation and fermentation of liquefied wheat bran starch to bioethanol was studied using coculture of amylolytic fungus and nonamylolytic sugar fermenting .

本工作致力于抗体片段纯化过程的多尺度优化。优化了生产过程中的色谱决策,包括色谱柱的数量及其大小,每批的循环数以及操作流速。使用基于微型实验数据的制造规模模拟数据集,建立了以负载质量、流速和柱床高度为输入的色谱通量数据驱动模型。与其他方法相比,分段线性回归建模方法具有简单、预测精度高的优点。提出了两种混合整数非线性规划(MINLP)模型,结合数据驱动模型,以最小化每克抗体纯化过程的总成本。然后,使用线性化技术和多参数分解将这些MINLP模型重新构造为混合整数线性规划(MILP)模型。研究了两个具有不同色谱柱尺寸替代品的工业相关案例,以证明所提出模型的适用性。
